Having routine (like regular time, and/or path) makes it easier for someone to kill you if they plan to.
Apart from that, if you're in some normal business - familiar route is good, but variety gives you extra experience and choices in case of road blocks, changes of traffic etc.
I mix it - shortest route is nice and safe, except during afternoon rushours, when I take a nicer, but slightly longer route.
