Tiagra will feel like a boat anchor, you'll have fewer gear inches for identical gears, and it'll rattle and jump like crazy.
Not really, I can't imagine a "feelable" difference.
I use SRAM cassette with my Ultegra setup and it's super quiet and shifts silky smooth.
