Why the crease in these handlebars?
Just curious if these creases serve a purpose or is it just some general design?

The handlebars were a (post crash?) replacement more recently, They are nor original.. yea, for aero brake levers the cable housing is laying in that groove..

As above, for aero cable routing. But, I've found that I prefer cables routed along the top front and rear of the bars to give a more comfortable hand grip.

Since I don't have the modern shifters, will it be awkward or uncomfortable using these handlers do you guys think ?
Maybe I could put an old cable in there for a filler ?

The best way is to fill it with auto body filler. Then cut a piece of wood with an arc to match the diameter of the handlebar plus the thickness of the sandpaper you are using. Sandpaper grit isn't super critical, 50, 100 and then 200 would probably be good enough. Then you can just tape it normally. If you just tape the handlebar without filling the groove first you might be able to feel the difference, but I doubt it.

Just leave it be. Doubt it will be noticed or felt after the bars are wrapped.
