The kind of caliper you can use is generally determined by the type of brake lever you have. Road brake lever needs a road caliper, flat bar MTB lever needs a MTB caliper

I don't think a lot of companies are using 20" wheels with disc brakes, but perhaps with some E-Bikes or folding bikes. Trikes?
Nonetheless, it would be easy enough to build a 20" wheel with a disc brake hub.
I think your leverage for stopping would be better with a 20" wheel than a 26"/29", so there would be no need to choose a very large disc.
As @Trevtassie mentioned, choose your calipers based on the handlebars and brake levers you choose.

I'd choose the smaller disc possible with a 20" tire. No need to have a 160 rotor on a 20".

You can easily build a 20" wheel with a disc brake hub. However, the calliper does not mount on the hub, it mounts on the frame. In order to use a disc brake 20" wheel your frame has to have mounting points for the calliper

Drum brakes have fallen out of vogue, but the mounts should be simpler.

I've seen several bikes and trikes with 20" disc. You will probably have to have the wheel built. Select hub, rim, spokes, rotor.

I have a 20"/406 wheel built on a disc hub and have used it with a disc brake a couple of times - both short trials- on the front of a long wheelbase recumbent in place of a 26"/559 wheel. The brake caliper doesn't care what size the wheel is. That's a good thing about disc brakes - different size wheels can be swapped in and out as long as the wheel fits the fork or rear of the frame.

20" wheels with disk brakes are very common on recumbent trikes. As others have said: it's relatively easy to have a wheel built with whatever hub and whatever rim you choose.

Since your screen name is "Ebikebeginner", you might be interested in this: https://rosecityroselandia.com/ It has 20" wheels and disk brakes.
It was designed and is being marketed by a couple friends of mine.
