Subtle But Persistent Bump On Rear Tire

So I have started riding a new bike that was put away for a while and I have found that there is a subtle bump coming from the rear tire. when dry spinning the tire it appears uneven but it wasn't a very solid test, so I took a closer look and i found on one spot where the tread was completely flat to the tire and on the rest of the tire it was fine. Could this one flat part be causing a bump or would it be that the rim is untrue.

Yes, the flat spot (caused by sitting in one place for a long time) is the likely cause. You may have to replace the tire. Long term storage should be with wheels off the ground.

It could be a flat spot from sitting a long time or it could be that the tire's not seated just right on the bead. Many tires have a thin molded "bead line" on the sidewall that should follow the outline of the rim. If your tire has such a line, take a look at it near your flat spot to see if it rises or dips. If it does, deflate the tire as completely as possible, then reinflate to re-seat it. Also make sure your tube isn't pinched in this area because that can keep the tire from seating properly.
