I am not aware nor have a read of an account of doing route des grandes alpes in "one go" although it is certainly achievable for a very select number of ultra endurance athletes. I've done the route fairly quick with a full loaded touring bike, traversing about 3 difficult passes per day. The route is about 450 miles a 5-60,000 feet of climbing with many, many Hors Category climbs. I know that many online routes indicate only 34,000 feet of climbing but I do not believe it unless they are just adding up the vertical from the 10-12 big climbs, in which case that sounds about right.
If your one go attempt will be w/o a support car, you really need to think and plan out your food for the southern middle of the route, IIRC. South of Briancon or something like that.

Just did the Route from South to North. It took 2,5 days - just about 60hours.
( 35 hours ride time)link to strava at the bottom. Decided to catch more sleep (4hours a day) and bit more stops to just enjoy the ride.
From the sport point of view - 50hours is achievable, but it wont be much enjoyable
