Windows 10 by default performs weekly defrags (or retrim for SSD), unless it was manually disabled or changed. At this point I'm suspecting either some recent Windows update screwing things up (more likely), or some hardware starting to go bad (less likely). That would make most sense and it is a work computer so updates have to get applied when they come out. It's possible that IT might have also put up some kind of security block on it, though I'm not sure why it wasn't considered a problem until now?
Going to try it again and if it still gives me problems I have a home computer I can run it from. It's an Acer Aspire computer, so not as good but should be fine to run Zwift.
