Strength and aerodynamics are the main benefits. There are drawbacks too. At some point deep rims become heavy, especially when made of aluminum. Crosswinds can be nasty with deep wheels, especially a deep front wheel.

If I've got a frame with 25.4/28.6 tubing and a threaded 1" steerer I'll stick with box section rims or rims less than 30mm deep if possible.
